Study on Meiotic Maturation of Mouse Oocyte in Vitro


Abstract
The expose of oocyte to spermatozoa before the completion of its maturation may result in fertilization failure, retarded and abnormal embryo growth, and reduced embryo viability.
To estabilish the possibility and extent of meiotic maturation of primodial mouse oocyte in vitro, the following study was performed.
1) Extrusion rate of the 1st polar body without PMSG in mice (1st group was 2.66¡¾3.57%). Inprimed PMSG for 24 hours(2nd groups), the rate was 4.72¡¾2.32%. and in primed PMSG for 48 hours(3rd group), 6.96¡¾3.4%.
2) After 12 hours of culture in vitro, the extrusion rate of 1st polar body in the 1st group was 5.74¡¾4.57%. In the 2nd group, the rate was 6.51¡¾4.4%. And in the 3 rd group, the rate was 10.12¡¾6.49%
3) After 24 hours of culturing in vitro, the extrusion rate of 1st polar body in the 1st group was 28.96¡¾7.03. In the 2nd group, the rate was 35.93¡¾13.93%. and in the 35.93¡¾13.93%. and in the 3rd group, the rate was 41.40¡¾13.96%).
4) After 48 hours of culturing in vitro, the extrusion rate of 1st polar body in the 1st group was 40.68¡¾10.94%. In the 2nd group, the rate was 52.24¡¾11.21%.And in the 4rd group, it was 56.24¡¾8.01%
5) There was a significant difference in the extrusion rate of 1st polar body culture in vitro between the 1st group and the 2nd group(P<0.05), and also the 2nd group and the 3 rd group(p<0.05)
6) In conclusion, the immature oocyte in vitro could be matured without the bormonal stimulation. However these oocytes could be matured more readily with the normonal hyperstimulaiton during the pre-ovualtory phase.
7) Therefore the IVF program, the immature oocytes sould be reduced along the pre-ovulatory oocytes. The successful in vitro maturation of these and should therefore increasethe oppotunity for securing a pregnancy.
